چکیده

Sermonti, G. (Istituto Superiore di Sanità, Rome, Italy), Milena Bandiera, and Isabella Spada-Sermonti. New approach to the genetics of Streptomyces coelicolor. J. Bacteriol. 91:384-392. 1966.-Mixed cultures of complementary auxotrophic strains of Streptomyces coelicolor A3(2) were preincubated on discs of cellophane on complete medium and were then transferred onto selective media. When one strain was streptomycin-resistant and the other was streptomycin-sensitive, and the transfer medium contained streptomycin, distinct minute tufts of aerial mycelium appeared on the background growth of the mixed culture. They turned out to be heterozygous clones (heteroclones) in which the streptomycin-sensitive allele was, as a rule, missing. The pattern of marker contribution of the streptomycin-sensitive parent to the zygotes was indicative of a continuous structure carrying the hereditary material. A gradual transfer of the donor genome during conjugation was suggested by the progressive completion of the zygotes obtained by increasing preincubation time.

A new kind of fertility variant in Streptomyces coelicolor.

class of fertility variants, called “ultra-fertile” (UF) , has been identified A in the actinomycete Streptomyces coelicolor A3 ( 2 ) (HOPWOOD, HAROLD, VIVIAN and FERGUSON 1968); these variants differ strikingly in their sexual capabilities from the strains that had previously been encountered in S. coielicolor genetics (HOPWOOD 1967). It seems likely that UF variants will contribute to an unde...

Genome sequence of a new Streptomyces coelicolor generalized transducing bacteriophage, ΦCAM.

Streptomyces coelicolor is a model system for the study of Streptomyces, a genus of bacteria responsible for the production of many clinically important antibiotics. Here we report the genome sequence of ΦCAM, a new S. coelicolor generalized transducing bacteriophage, isolated from a soil sample originating from Lincolnshire, United Kingdom. Many open reading frames within ΦCAM shared high leve...

A recA null mutation may be generated in Streptomyces coelicolor.

The recombinase RecA plays a crucial role in homologous recombination and the SOS response in bacteria. Although recA mutants usually are defective in homologous recombination and grow poorly, they nevertheless can be isolated in almost all bacteria. Previously, considerable difficulties were experienced by several laboratories in generating recA null mutations in Streptomyces, and the only rec...

Region-specific transcriptional activity in the genome of Streptomyces coelicolor A3(2).

Transcriptional analysis of microbial genomes is an important component of functional genomics. Strategies such as hybridization of labeled total RNA against ordered clone libraries or differential-display approaches have already been carried out to identify expressed genes. We describe here an additional method which applies subtractive hybridization between genome-specific DNA and total RNA f...

The Streptomyces coelicolor A3(2) lipAR operon encodes an extracellular lipase and a new type of transcriptional regulator.

A region of the Streptomyces coelicolor A3(2) chromosome was identified and cloned by using as a probe the lipase gene from Streptomyces exfoliatus M11. The cloned region consisted of 6286 bp, and carried a complete lipase gene, lipA, as well as a gene encoding a transcriptional activator (lipR). The S. coelicolor A3(2) lipA gene encodes a functional extracellular lipase 82% identical to the S....
